Quantitative spectroscopy of single molecule interaction times

چکیده
Single molecule fluorescence tracking provides information at nanometer-scale and millisecond-temporal resolution about the dynamics interaction of individual molecules in a biological environment. While dynamic behavior isolated can be characterized well, quantitative insight is more limited when interactions between two indistinguishable occur. We address this aspect by developing theoretical foundation for spectroscopy times, i.e., inference from imaging data. A non-trivial crossover power law to an exponential distribution times highlighted, together with dependence term upon microscopic reaction affinity. Our approach validated simulated experimental datasets.

Force spectroscopy of barnase-barstar single molecule interaction.
Results of the single molecule force spectroscopy study of specific interactions between ribonuclease barnase and its inhibitor barstar are presented. Experimental data obtained for the force loading rate ranging 2-70 nN/s are well approximated by a single straight line, from which the dissociation barrier of the width of 0.12 nm and height of 0.75-0.85 × 10(-19)J can be inferred. The measured ...
متن کاملQuantitative analysis of single-molecule RNA-protein interaction.
RNA-binding proteins impact gene expression at the posttranscriptional level by interacting with cognate cis elements within the transcripts. Here, we apply dynamic single-molecule force spectroscopy to study the interaction of the Arabidopsis glycine-rich RNA-binding protein AtGRP8 with its RNA target. A dwell-time-dependent analysis of the single-molecule data in combination with competition ...
متن کاملSingle-photon spectroscopy of a single molecule.
Efficient interaction of light and matter at the ultimate limit of single photons and single emitters is of great interest from a fundamental point of view and for emerging applications in quantum engineering. However, the difficulty of generating single-photon streams with specific wavelengths, bandwidths, and power as well as the weak interaction probability of a single photon with an optical...
متن کاملSingle-molecule spectroscopy and microscopy
Advances in detector sensitivity and improvements in instrument design have recently provided scientists with tools to probe single molecules with light, and monitor their photophysical properties with exquisite sensitivity and spatial as well as temporal resolution. Spectroscopic and temporal information is used to explore molecular structure, conformational dynamics, local environment and int...
متن کاملBroadband single-molecule excitation spectroscopy
Over the past 25 years, single-molecule spectroscopy has developed into a widely used tool in multiple disciplines of science. The diversity of routinely recorded emission spectra does underpin the strength of the single-molecule approach in resolving the heterogeneity and dynamics, otherwise hidden in the ensemble. In early cryogenic studies single molecules were identified by their distinct e...
